Officers swooped on four males at the airport, where the teenage murder suspect was travelling under a false name, police allege.
The 25-year-old was found in the middle of the road at North Terrace, suffering multiple stab wounds to his chest.
He died shortly after police arrived at the scene.
The other three males police detained at the airport – aged 16, 18 and 21 – have been charged with aggravated affray.
The 16-year-old will appear in the Adelaide Youth Court today, while the other three have been refused bail and will front Adelaide Magistrates Court, also today.
SA Police believe the alleged stabbing is gang-related.
A search for suspects saw multiple people arrested and a machete seized.
Star Group officers from the Quest Hotel on Franklin Street hauled seven people from their rooms, as detectives searched for evidence.
The alleged stabbing is one of several fights that broke out between the suspects last night.
Police are appealing for anybody who witnessed any violent behaviour to come forward and assist with investigations.
